Lipid nanoparticles (LNPs) are a promising drug delivery system composed of a mix of various lipids (e.g., helper, ionizable, PEGylated, targeting) and often cholesterol with an encapsulated cargo. We offer a workflow solution to optimize LNP formulations, utilizing automation and ultracentrifugation, as well as solutions of biophysical and function studies with analytical ultracentrifugation and flow cytometry.
LNP Workflow
Drug Development
Therapeutics, including small molecules (e.g., antibiotics, chemotherapy agents), biologics (e.g., antibodies, hormones), and nucleic acids (e.g., mRNA, siRNA), are prepared using ready-to-use reagents, liquid handlers, automated cell culture workstations, centrifuges, and chromatography systems.
Lipid Nanoparticle Production
LNP formulations incorporating novel ionizable cationic lipids are at the forefront of RNA delivery technologies. Encapsulation involves using a microfluidic mixer to facilitate the controlled nanoprecipitation of lipids and mRNA in an acidic buffer through electrostatic interaction.
Final Preparations
Following encapsulation, buffer exchange is performed to facilitate the rearrangement and stabilization of the nanoparticles. Fractionation of LNPs with different loading levels is also critical and can be achieved with centrifugation to ensure optimal transfection potency.
Analysis & Characterization
Characterizing LNP attributes, such as LNP size, encapsulation efficiency, drug purity, heterogeneity, and mRNA/siRNA loading levels, before in vitro or in vivo use is essential for reproducibility and can be achieved with the Optima AUC Analytical Ultracentrifuge, LS 13 320 XR Particle Size Analyzer, and CytoFLEX Flow Cytometers.
