Single Laser Line Excitation, Negligible Emission Overlap
In contrast to the vast majority of fluorescent dyes used in flow cytometry, infrared fluorochromes IR820 and IR870 are mainly excited by the infrared laser, hence emitting in the infrared range with practically no overlap with other fluorochrome emissions.
Ultralow Autofluorescent Background Is Inherent To IR Laser Excitation
Autofluorescence presents significant challenges in flow cytometry, particularly in spectral flow cytometry, complicating the detection, quantification and interpretation of fluorescence signals. IR820 and IR870 signals are nearly devoid of autofluorescence by design, as they are excited and fluoresce in the infrared range.
Infrared Fluorochromes IR820 and IR870 - Moderate Bright Signals, Suited For Gating Markers
IR820 and IR870 conjugates deliver signals which match the brightness needed for clear separation of gating makers, but their emissions are still moderately bright so they do not cause mutual spillover spreading.

No undesired dye-dye interactions could be observed for the CD20-IR820 conjugate when used within a 21-color panel. Data was acquired from stained and lysed whole blood on a CytoFLEX LX flow cytometer equipped with a CytoFLEX mosaic 88 spectral detection module.

CD45-IR820 could be used together with the infrared fixable viability dye ViaKrome 808 to clearly identify viable human leukocytes in the sample. Data was acquired from stained and lysed aged whole blood on a CytoFLEX LX.
