CytoFLEX nano: Calibrating Light Scatter Data to Diameter using FCMPASS™
Objective:
This document provides guidance on calibrating arbitrary units of Violet Side Scatter (VSSC) data from the CytoFLEX nano flow cytometer into standard units of diameter using FCMPASS™ software. The primary objective of light scatter calibration is to enable instrument agnostic comparisons of data by utilized standard units. An additional benefit of utilizing standard units, such as size, are the biological insights their characterization provides.
In this application note you will learn:
-
The importance of calibrating arbitrary units to standard units in nanoparticle research.
-
How to calibrate the CytoFLEX nano VSSC1 parameter’s data in diameter units of nanometers
by utilizing an approximate refractive index (RI), calibration reference particles with a known
size and RI, and using a third-party Mie-modeling software; FCMPASS™. -
How to import FCMPASS™ calibrated .fcs files into the CytExpert nano software
